The home side were 1-0 down in the second half of Tuesday's third-round tie when they looked to launch an attack from the half-way line.
But unfortunately, referee Andy Davies was in the way of the pass and deflected the ball away from the Reading players and into the path of new Swansea signing Renato Sanches.
He played in Ayew, who controlled smartly before cutting inside and producing an emphatic finish into the bottom right corner - followed by a chorus of boos by the home fans.
Swansea held on to win 2-0 and progress into the third round of the Carabao Cup.
