The Rams, parading a team which featured 11 changes from the one which thumped Hull 5-0 in the league last Friday, began positively.
Barnsley nearly took the lead when Harvey Barnes smashed a pile-driver against the crossbar from distance.
But the Rams went in front after six minutes.
Chris Martin teed up Johnny Russell to curl home into the corner from 12 yards.
It was end-to-end stuff with Barnsley’s Tom Bradshaw missing a sitter from close range after Jonathan Mitchell parried from Jason McCarthy.
Barnes went close again minutes later when his half-volley was deflected behind off Andre Wisdom.
It was even on 18 minutes when Adam Jackson rose to head his first Tykes goal from a Ryan Hedges corner.
But back came the Rams — they ought to have re-taken the lead immediately when Russell side-footed straight at Davies from ten yards.
And eight minutes before the break, Derby threatened again.
Mason Bennett hung up a terrific cross for Ikechi Anya but his header was easy for Davies.
Anya turned provider 60 seconds later when Bennett tucked home his far-post cross to put Gary Rowett’s side back in front.
Brad Potts had a gilt-edged chance to equalise just before the break.
The Tykes midfielder headed miserably wide from six yards when it looked easier to score.
Despite the complete change of line-up, Rowett’s Rams had been good value for their lead at the break and also opened the second period impressively.
Tricky left-footer Bennett hammered in a shot from the edge of the box that Tykes keeper Davies had to dive full length to push just round the post.

Barnsley responded, with Potts seeing a crisp drive deflected and parried by Derby stopper Mitchell.
With an early-season visit to Wembley still very much up for grabs, the Rams went agonisingly close to stretching their lead just before the hour mark.
Again Wisdom swung over a tempting cross, with Bennett’s pinpoint header tipped brilliantly over the top by the superb Davies.
It was Rowett’s makeshift Rams looking the most likely to be facing Spurs — who are playing their home games at Wembley this season while their White Hart Lane home is redeveloped.
But the Tykes were not out of it and Matty Pearson failed to make the Derby pay when he blazed a shot wildly over the top.
The visitors did not heed to warning and Barnsley levelled on 74 minutes when Tom Bradshaw glided in to convert a fizzing low cross from Barnes.
Sub Hammill then sealed the hosts’ dream Wembley trip with his jaw-dropping strike in the 88th minute.
